In Vitro Endothelialization of PTFE Vascular Grafts : A Comparison of Various Substrates, Cell Densities and Incubation Times

Afin de déterminer in vitro les conditions optimales d’endothélialisation des prothèses vasculaires de petit calibre, l’adhésion des cellules endothéliales (ECs) cultivées à partir de veines ombilicales humaines étaient étudiées à la surface interne de prothèses en polytétrafluoroéthylène (PTFE). L’attachement et la prolifération cellulaire étaient comparés sur des prothèses en PTFE ayant (a) été traitées préalablement par de la fibronectine (HFN), du collagène de type I, du collagène de type IV, du plasma, et de la colle biologique avec ou sans thrombine, seul ou en combinaison; (b) ensemencées avec des densités cellulaires variant de 0,5 x 105 à 6 x 105 cellules/cm2; et (c) incubées pendant 30, 60 ou 90 min. L’attachement et la prolifération cellulaire étaient estimés par microscopie électronique à balayage. Le recouvrement quantitatif des cellules endothéliales à la surface des prothèses était évalué par un analyseur d’images. Pour déterminer les conditions optimales d’endothélialisation parmi les 189 traitements possibles, nous avons utilisé l’analyse de la variance. En conclusion, une monocouche de cellules confluentes peut être en pratique obtenue sur des prothèses en PTFE de petit calibre quand celles-ci sont pré-traitées avec de la colle biologique ou du plasma, ensemencées avec des densités cellulaires ≥4 x 105 cellules/cm2, et incubées pendant 60 min. Tous ces paramètres sont compatibles avec une intervention chirurgicale.
Summary
To establish the optimal conditions for achieving endothelial cell coverage of the luminal surfaces of small-caliber vascular grafts in vitro, the attachment of endothelial cells (ECs) cultured from human umbilical veins to polytetrafluoroethylene (PTFE) grafts was studied. Cell attachment and spreading were compared after PTFE grafts were (a) precoated with fibronectin (HFN), type I collagen, type IV collagen, plasma and fibrin with or without thrombin, singly or in combination; (b) seeded with cell densities varying from 0.5 x 10(5) to 6 x 10(5) cells/cm2; and (c) incubated at 30, 60, or 90 min. Cell coverage and spreading were assessed by means of scanning electron microscopy. Quantification of graft surface coverage was performed with computer-assisted image analysis. To determine optimal conditions of endothelialization among the 189 treatment combinations, analysis of variance was used. We conclude that a virtually confluent cell monolayer can be established on small-caliber PTFE grafts when precoated with fibrin glue or plasma, seeded with cell densities >/=4 x 10(5) cells/cm2, and incubated for 60 min. These parameters are compatible with an operating room vascular procedure.
